Mutations in the Fc-region of IgG from synovial fluids of patients with rheumatoid arthritis

چکیده
Rheumatoid arthritis an antigen-driven autoimmune disease and the eliciting antigens are unknown. They should be present in immune complexes of synovial fluids. The immune complexes were isolated from synovial fluids of patients with rheumatoid arthritis and analyzed by polyacrylamide gel electrophoresis and Western blotting. Immune complexes contained IgG as major protein. The antibodies dissociated at pH 2.5 and self-aggregated upon neutralization. In ELISA assays, they had the highest specificity to homologous IgG, less to normal IgG and lowest of heterologous arthritic IgG confirming earlier results that altered IgG structures were antigenic. Sequencing of the Fc-regions revealed mutations in all patients analyzed. Therefore, mutations within the Fc-regions of IgG could be antigens in rheumatoid arthritis. We propose that methods should be developed to prove this hypothesis. Subjects: Biochemistry; Immunology; Rheumatology

Comparative Measurement of Rheumatoid Factor in Serum and Synovial Fluid of Rheumatoid Arthritis Patients by ELISA and Latex-Agglutination Test
Rheumatoid factors (RF) are autoantibodies with specificity for the Fc portion of human IgG. Production of RF is a characteristic feature of rheumatoid arthritis (RA) and is detectable in high titer in about 90% of these patients. In this study, we measured total IgM, IgA, IgMRF and IgARF in serum and synovial fluid of 45 RA patients by ELISA and the results were compared with those obtained b...
متن کاملInhibition of Fc-rosette formation by synovial fluids from patients with rheumatoid arthritis.
Human red cells sensitized with one of the Rh antisera (Ripley) form rosettes (Fc-rosette) with human B lymphocytes and the rosettes are well inhibited by aggregated human IgG. As synovial fluids from patients with rheumatoid arthritis are usually hypocomplementaemic and frequently contain immune complexes, they were used for the inhibition of Fc-rosette formations in this study. The Fc-rosette...

Background: Heat-shock proteins are part of a strictly controlled biological system that allows organisms to respond to environmental stresses. Different proinflammatory cytokines are present in the synovial tissue of rheumatoid arthritis patients. Such tissues respond to stress and induce heat-shock proteins. In addition, synovial cells are exposed to mechanical stress caused by joint motion. ...
متن کاملIgG rheumatoid factor synthesis in synovial fluid.
Sir: Winchester et al two decades ago proposed that IgG rheumatoid factor (RF) could be locally produced in plasma cells that line the synovium.' We conducted a short study looking at paired samples of synovial fluid (from the knee joint) and serum from patients with definite rheumatoid arthritis to determine if the production of IgG RF is localised in the synovial fluid. Thirty six serum sampl...
متن کاملDefining the genetic origins of three rheumatoid synovium-derived IgG rheumatoid factors.
A major diagnostic marker in most rheumatoid arthritis (RA) patients is the rheumatoid factor (RF), an autoantibody that binds to the Fc region of IgG. To delineate the Ig genes and the underlying mechanism for RF production in RA patients, we applied a systematic approach to define the genetic origins of three IgG RFs derived from the synovial fluid of two RA patients. The results show that tw...
